In 2004, Guinoo joined the popular reality search Star Circle Teen Quest via ABS-CBN and made it to the Top 5 finalist or much known as the "Magic Circle of 5", along with co-questorsHero Angeles, Sandara Park, Joross Gamboa, and Melissa Ricks. On June 2004, the Grand Questor's Night was held in Araneta Coliseum, and she won 2nd Runner up. She was launched as one of the newest member of ABS-CBN's Star Magic. She was also paired, as a love team, with Joross Gamboa, even before Star Circle Quest ended. Her first show withABS-CBN was with the youth oriented drama SCQ Reload, with co-former SCQ finalists.

She also played in movies like Now That I Have You , Can This Be Love, and D' Anothers.

Her biggest break came when she was one of the lead casts of the drama series Sineserye Presents: Natutulog Ba Ang Diyos?, with big stars like Dina Bonnevie and Rosanna Roces. In 2008, she started her first-ever solo teleserye in ABS-CBN entitled Ligaw na Bulaklak which airs in an afternoon slot. Presently she is one of mainstay of comedy show entitle Everybody Hapi over TV5 as Jenny with John Estrada, Eugene Domingo, Long Mejia, Alex Gonzaga and Matt Evans.

She is half German. She is happily married to Elton Yap and have a beautiful daughter name Rain Eliana Guinoo-Yap. Elton and Roxanne got married this year on January 23, 2011. The same as the wedding date of one of showbiz couples Gladys Reyes and Christopher Roxas.

Before she got married, she had dated her former loveteam for 3 years Joross Gamboa. Their loveteam was called "JoRox" and their fans club was called "KULTONG JOROX". They were together for 2 years from 2004-2006. In 2007, she dated Jake Cuenca. They were teamed up after Roxanne and Joross separated. They're first team up was in Sineserye Presents: Natutulog Ba Ang Diyos? love triangle with Gamboa. They were together for 1 year. Their relationship was a controversy. Allegations said that Cuenca had hurt Guinoo when they were in Guam (people saw her bruises on her arm) due to their show called ASAP. Neither parties denied nor confirmed the issued.

She was on the top of her career when last February 2010 she confirmed that she was 2 months pregnant to her boyfriend then Elton. They cannot get married last year due to Elton's sister who got married that same year (January 2010). Roxanne was interviwed on the BUZZ by Boy Abunda last year and confirmed that she was pregnant to her boyfriend who was 2 years younger than her. In September 2010, she gave birth to a beautiful baby girl that they named Rain Eliana a combination of the names of their mothers. Eli (Elisabeth, Elton's Mother) and ana (Roxanne's mother), Rain was from her name Roxanne. She promised that she will not leave the showbusiness but she has to concentrate first to her family.
